Offer DescriptionThe project aims to develop a model of personalized treatment for aggressive CTCL patients by the integration of flow-cytometric and immunohistochemical data with the disease outcome and response to treatment.
IHC analysis of CD39 and CD73 expression in skin infiltrating cells and local microenvironment (ME) of CTCL.
Neoplastic cells and TME will be analyzed by IHC and/or FACS in pre-and post-treatment intralesional skin biopsies collected at diagnosis and/or at follow-up in the SS cohort described above (Aim 3B) and in a retrospective MF cohort diagnosed and followed-up.
IHC will be used to evaluate CD39 and CD73 expression along with the diagnostic T-cell markers and molecules involved in the TME modulation such as TIM-3, PD-1, and LAG-3.
Data obtained for aggressive MF (about 30%) will be compared to those of indolent MF.
Fresh skin biopsies will be minced (mechanically and enzymatically) and analyzed by FACS.
