Offer Description Azathioprine (AZA) is used to treat chronic inflammatory bowel disease (IBD) and may cause severe adverse effects in patients with early-onset disease (VEO-IBD).

Inter-individual differences in thiopurine methyltransferase (TPMT) activity, which is higher in patients with VEO-IBD, could explain the altered response to AZA.

The PACSIN2 rs2413739 variant has been associated with TPMT activity in leukaemic patients, so its effect on TPMT activity has also been hypothesised in patients with VEO-IBD.

Epigenetic factors could also explain the altered TPMT activity as it changes with age.

This study will evaluate the role of PACSIN2 rs2413739 and TPMT gene methylation as markers of AZA efficacy and create in vitro models to study these associations.

In addition, new pharmacogenetic markers obtained by next-generation sequencing will be explored.
