Offer Description The project will focus on studying the role of ER-phagy in modulating phenotypic severity in the disease Osteogenesis Imperfecta (OI). OI, the most common among hereditary bone diseases, is caused by dominant mutations in the COL1A1 and COL1A2 genes which encode the a chains of collagen I. These mutations cause partial retention of the mutated protein in the endoplasmic reticulum (ER). A type of autophagy, defined as ER-phagy, appears to be involved in reducing the accumulation of poorly folded proteins in the ER, including collagen. The aim of the project will be to analyze the role and in vivo regulation of ER-phagy in the clearance of misfolded collagen I molecules in OI.
Animal models of OI will be used to study and identify key regulators of ER phage processing and its contribution to cellular responses associated with intracellular accumulation of collagen I.
Main Requirements Degree (with a course duration of not less than 4 years, as referred to in the pre-existing academic regulations pursuant to D. M. n. 509/1999) or post-graduate degrees (pursuant to D. M. n. 509/1999 and to n. 270/2004) awarded by Italian or foreign Universities recognized as equivalent.
Scientific or professional curriculum suitable for conducting the research activity.
If available, any Ph. D. title (Art 2- par. 1).
Further Specific Assessable Qualifications Proven experience of cell cultures of eukaryotic cells
Proven experience in the use of biochemical and molecular techniques (including but not limited to western blotting, real-time PCR)
Proven experience in the use of histological techniques on tissues (tissue preparation, embedding, sections and stains or immunohistochemistry)
Proven experience in the use of fluorescence microscopy (confocal and super-resolution)
Proven experience in the use of animal models
Excellent knowledge of the English language
International publications on topics relating to the characterization of animal models of skeletal diseases
