Mitochondria As A Novel Therapeutic Target In Fragile X-Related Disorders

ALMA MATER STUDIORUM - UNIVERSITA' DI BOLOGNA - DIPARTIMENTO DI SCIENZE BIOMEDICHE E NEUROMOTORIE Job Overview Recent data indicate that mitochondrial function is deranged in Fragile X-related Disorders (FXDs).

Our findings show upregulation of several mitochondrial proteins in fibroblasts derived from FXS, FXTAS patients as well as UFM carriers, including antioxidant enzymes, OXPHOS complexes and cyclophilin D (CypD), the main modulator of the mitochondrial permeability transition pore (PTP) inducing apoptosis.

These findings suggest novel mitochondrial mechanisms that may be responsible for the pathological processes in FXDs and can be targeted through new pharmacological treatments.

Through NMR spectroscopy, biochemistry, proteomics and pharmacological approaches we aim at defining a novel therapeutic approach for FXDs.

Finally, the iPSCs technology will allow to reprogram patient cells and differentiate them into neurons, validating both the dysregulated pathways identified in fibroblasts, and the pharmacological compounds rescuing the FXD phenotypes.
